‘欧美杨107’ 组培苗瓶外生根

摘 要:

‘欧美杨107’是近年来重点推广的杨树品种之一, 但其组培苗存在移栽成活率不高的问题。本试验研究了‘欧美杨107’组培苗的直接瓶外生根和瓶内复壮后瓶外生根两种生根方式, 结果表明, 两种方式蘸取IBA的最佳浓度分别为10 mg•L-1和100 mg•L-1, 生根率分别达73.33% 和81.67%, 移栽成活率分别为62.5% 和93.33%。进一步研究发现生根的过程中外源补加0.5 mg•L-1 IBA 溶液能加速生根, 使生根周期缩短2 周左右。添加 1/2MS 营养液可促进生根苗茎的生长和根的伸长, 有利于 移栽成活。瓶内生根苗的生根率高达到95%, 但是移栽难以成活, 显微镜观察发现瓶外生根苗比瓶内生根苗的根毛发达, 验证了瓶外生根苗的根吸收功能更好。该研究对促进‘ 欧美杨107’ 组培苗及其转基因苗的推广应用具有重要的现实意义。

Ex vitro Rooting of Populus×euramericana cv. Neva Microshoots

Abstract:

Populus×euramericana cv. Neva is one of the most widely used species in recent years, but its low transplant-survival rate of in vitro rooted plantlets leads to limited application. We investigated ex vitro rooting of Populus×euramericana cv. Neva in two ways, direct ex vitro rooting of microshoots and ex vitro rooting of rootless microshoots pre-cultured in root induction media. The optimal concentrations of IBA were 10 mg·L-1and 100 mg·L-1 for the two methods, respectively. The rooting percentage was 73.33% and 81.67%, transplantsurvival rate reached to 62.5% and 93.33%. Further, in the rooting process, rooting period was shorten by two weeks by adding 0.5 mg·L-1 IBA, and elongation of stems and roots were promoted by adding 1/2MS nutrient liquid. Rooting percentage of in vitro rooted plantlets reached up to 95%, but its transplant-survival rate was low. We found that ex vitro rooted plantlets had more root hairs than those of in vitro rooting, which resulted in better absorption. This study offers potential benefits in spread and application of test-tube plantlet of Populus×euramericana cv. Neva.
